In this work, a hydro-alcoholic extract of manna (HME) from Sicilian Fraxinus angustifolia Vahl was investigated using HPLC-DAD to find phenol components and using chemical and biological in vitro assays to determine its reducing, antioxidant and anti-inflammatory capacity. In several species of the Oleaceae, mannitol, already present at considerable levels, accumulates in response to stress. This family comprises both deciduous and evergreen species, and we investigated the role of mannitol in deciduous malacophyll and evergreen sclerophyll species growing under the same conditions in the field. The relationship between mannitol content and changes in rainfall or temperature was also studied. The mannitol content of leaves of Fraxinus ornus L., F. angustifolia Vahl., Olea europaea L. and Phillyrea media L. was determined by gas chromatography. Fraxinus angustifolia is used for afforestation and for production of both valuable timber and manna , a substance with pharmacological applications. The aim of this research was to establish the optimized condition for cryopreservation of F. angustifolia seeds, in order to protect the germplasm of this species. Germination percentage and mean time of germination of non-treated seeds (control) and liquid nitrogen-treated seeds were established in water or in 10 -6 M gibberellic acid (GA 3 ). The manna is a product obtained from the elaborate sap solidification that comes out from the incisions made on the stem of some species of the genus Fraxinus L. In this study, we have analyzed 12 samples of manna to determine the chemical composition. Twenty elements (Al, As, Ba, Cd, Co, Cr, Cu, Fe, Li, Mn, Mo, Ni, Pb, Rb, Sb, Se, Sr, U, V, Zn) were determined using an Inductively Coupled Plasma Mass Spectrometer. The results show that the most abundant elements are Fe > Rb > Al > Cu > Ba > Zn > Sr with median concentrations in the range of 0.9 – 10mg kg-1. Fraxinus angustifolia subsp. angustifolia is a plant with an age-old use for the production of manna. However, it is also a valuable source of fixed oil rich-seeds. In the present study we examined the chemical and biological properties of this oil in order to support a possible application in foodstuffs, nutraceuticals and cosmetics. ABSTRACT The content of mannitol and malate was assayed enzimatically during spring, summer and autumn, in leaves of two species of ash, Fraxinus ornus L. and Fraxinus angustifolia Vahl, traditionally cultivated in Sicily for the extraction of manna. Both species contain high levels of mannitol and show, on a dry weight basis, a 65–80% increase in the summer content of this polyol. The malate content differs in the two species: in F. ornus it shows a summer increase, but it is relatively low (65–115 µmol g-1 DW), while in F. angustifolia it is higher (275–318 µmol g-1 DW), but remains more or less constant throughout the year.
